SS-31 (Elamipretide / MTP-131 / Bendavia) is a synthetic mitochondrial-targeting tetrapeptide developed by the Hazel Szeto laboratory and licensed to Stealth BioTherapeutics. The peptide consists of an aromatic-cationic sequence (D-Arg-2β²,6β²-dimethylTyr-Lys-Phe-NHβ) designed to selectively concentrate in the inner mitochondrial membrane through electrostatic attraction to the negatively charged headgroup of cardiolipin β a unique mitochondrial phospholipid critical for organization of electron transport chain supercomplexes. By binding cardiolipin, SS-31 is studied for stabilizing the structural organization of the inner mitochondrial membrane, preserving cristae architecture, reducing reactive oxygen species (ROS) leak, and supporting ATP synthesis efficiency.
