Description
Dihexa (PNB-0408) is a synthetic oligopeptide derived from angiotensin IV, studied as a potent activator of the hepatocyte growth factor / c-Met receptor signaling system. The compound binds HGF with sub-nanomolar affinity (Kd ≈ 65 pM) and is investigated as a synaptogenic research molecule with notable properties unusual for a peptide: oral bioavailability and blood-brain barrier permeability. Dihexa has been studied in preclinical models of cognitive function, Alzheimer’s-like neurodegeneration, and dendritic spine formation, where it has been reported to promote functional synaptogenesis with high potency in neurotrophic assays. Originally engineered at Washington State University by the Harding lab and later licensed to M3 Biotechnology, Dihexa retains the angiotensin IV pharmacophore for AT4 receptor binding while incorporating N-hexanoyl and aminohexanoic acid modifications that confer metabolic stability and lipophilic BBB penetration. Its small molecular weight (~505 Da) and lipophilic character support oral capsule administration consistent with the published research literature.
