Description

TB-500 (commonly used as the product name for Thymosin Beta-4 / TΞ²4) is a naturally occurring 43-amino-acid peptide first identified in calf thymus extracts in the 1960s and characterized as a major intracellular actin-sequestering protein. Thymosin Beta-4 is the most abundant member of the Ξ²-thymosin family in mammalian cells and functions primarily through G-actin binding, regulating actin polymerization dynamics critical for cell migration, cytoskeletal remodeling, and tissue organization. The peptide has been investigated in preclinical research models for its effects on wound healing, angiogenesis, fibroblast and endothelial cell migration, anti-inflammatory signaling, and cardiac repair. Research interest extends to corneal and dermal repair, ischemic injury recovery, and the bioactive N-terminal tetrapeptide fragment (Ac-SDKP) released through TΞ²4 proteolysis. TB-500 is one of the most extensively studied actin-binding peptides in regenerative biology.

Benefits (Research Focus)

β€’ Actin sequestration β€” studied as the most abundant intracellular G-actin binding protein in the Ξ²-thymosin family

β€’ Cell migration research β€” investigated for fibroblast, endothelial, and stem cell migration via cytoskeletal reorganization

β€’ Angiogenesis β€” explored for VEGF expression and new blood vessel formation in healing tissues

β€’ Tissue recovery models β€” examined for cardiac, corneal, and dermal repair endpoints in preclinical research

β€’ Anti-inflammatory signaling β€” researched for balanced inflammatory and reparative pathway modulation

What Researchers Look At

β€’ G-actin binding affinity and actin polymerization dynamics in cell-based assays

β€’ Endothelial cell migration, tube formation, and angiogenesis markers (VEGF, integrins)

β€’ Cardiac repair and ischemic injury recovery endpoints in rodent models

β€’ Corneal and dermal wound closure rates in preclinical recovery research

β€’ Ac-SDKP tetrapeptide fragment formation and downstream anti-fibrotic signaling
